چکیده
Genome-wide association studies have previously identified 23 genetic loci associated with circulating fibrinogen concentration. These studies used HapMap imputation and did not examine the X-chromosome. 1000 Genomes imputation provides better coverage of uncommon variants, and includes indels. We conducted a genome-wide association analysis of 34 studies imputed to the 1000 Genomes Project reference panel and including ∼120 000 participants of European ancestry (95 806 participants with data on the X-chromosome). Approximately 10.7 million single-nucleotide polymorphisms and 1.2 million indels were examined. We identified 41 genome-wide significant fibrinogen loci; of which, 18 were newly identified. There were no genome-wide significant signals on the X-chromosome. The lead variants of five significant loci were indels. We further identified six additional independent signals, including three rare variants, at two previously characterized loci: FGB and IRF1. Together the 41 loci explain 3% of the variance in plasma fibrinogen concentration.

Genome-wide meta-analysis identifies new susceptibility loci for migraine
Migraine is the most common brain disorder, affecting approximately 14% of the adult population, but its molecular mechanisms are poorly understood. We report the results of a meta-analysis across 29 genome-wide association studies, including a total of 23,285 individuals with migraine (cases) and 95,425 population-matched controls. We identified 12 loci associated with migraine susceptibility ...
متن کاملIsolation and characterization of 13 new microsatellite markers in the triangle mussel (Hyriopsis cumingii)
Microsatellite enriched library of Hyriopsis cumingii was constructed according to the strong affinity between biotin and streptavidin. One hundred clones with foreign inserts were sequenced and 65 clones were screened for usable microsatellites, of which 36 were deemed unique, of sufficient length (more than 8 repeats) and possessed adequate flanking regions for primer designment. Among 36 pri...
متن کاملLarge-scale association analysis identifies 13 new susceptibility loci for coronary artery disease.
We performed a meta-analysis of 4 genome-wide association studies of coronary artery disease (CAD) comprising 22,233 individuals with CAD (cases) and 64,762 controls of European descent followed by genotyping of top association signals in 56,682 additional individuals. This analysis identified 3 loci newly associated with CAD at P < 5 × 0−8 and confirmed the association of 0 of 2 previously rep...
متن کاملMultiethnic meta-analysis of genome-wide association studies in >100 000 subjects identifies 23 fibrinogen-associated Loci but no strong evidence of a causal association between circulating fibrinogen and cardiovascular disease.
BACKGROUND Estimates of the heritability of plasma fibrinogen concentration, an established predictor of cardiovascular disease, range from 34% to 50%. Genetic variants so far identified by genome-wide association studies explain only a small proportion (<2%) of its variation. METHODS AND RESULTS We conducted a meta-analysis of 28 genome-wide association studies including >90 000 subjects of ...
متن کاملMolecular study of a consanguineous family with autosomal recessive mental retardation and speech disorder
Mental retardation (MR) is one of the most frequently found major genetic disorders around the world, affecting 1-3% of the people in the general population. The recent advancement in molecular biology and cytogenetic study has made possible the identification of new genes for a variety of genetic disorders including autosomal recessive MR. Recessive genetic disorders are common in Pakistan due...
